|Title:||Analysis Of An Adr1p Suppresser: Evidences For Regulation Through A Molecular Adapter|
|Abstract:||Adrlp is an activator involved in the derepression of ADH2 and genes encoding peroxisomal proteins in Saccharomyces cere.visiae. Recently, we showed that MOX, a specialized Hansenula polymorpha gene encoding the peroxisomal rnethanol oxidase, is regulated by Adrlp in S. ce.re.visiae (Pereira and Hollenberg, Eur. Jour. Biochem, 238: 1KI 191,1996). We found that this regulation is mediated by the binding of two Adrlp monomers to a MOX promoter re gion, named MOX-B. Remarkably, MO.Y-B mediated activation by Adrlp only when placed in the correct orientation. When inverted, the region was insensitive to Adrlp, though this activator may bind to the promoter region under this condition. We found a mutant which partially suppress the need of Adrlp for MOX derepression and in which Adrlp is able to activate the inverted :VfOX-B region. This suggests that the mutation may affect factors that bridge regulators and the basic transcriptional machine. To test this hypothesis wo are analyzing the effect of the mutation on the transcriptional ability of each Adrlp activation domain. The mutation also lead to overexpression of other promoters repressed by glucose and to flocculation, a feature related with th'1 mutation of several transrriptional repressers, like tupl, ssn6 and rox-3.|
